Chapter 51 Junior Sister, Senior Brother Will Go First

Chapter 51 Junior Sister, Senior Brother is Taking His Leave First (Please add to your favorites and read on!)
To be honest, the Medical Basic Cell Experiment Technique Kit is really useful. It includes all the procedures for common cell experiments.

After all of them were upgraded to Level 2, Fang Ziye's proficiency in the corresponding areas was at least no less than that of a typical mature academic master's student.

Let's use this analogy.

Fang Ziye worked diligently in the training room for two years, but his basic medical operation skills only reached level 2. Similarly, a level 2 proficiency in basic medical cell laboratory technology is equivalent to the proficiency achieved by an academic master's degree holder who has diligently cultured cells in the cell room for two years.

Yuan Weihong followed a professional graduate program, pursuing both a professional master's and a professional doctoral degree. In reality, he didn't spend much time in the lab, so his proficiency level wasn't very high.

Fang Ziye went downstairs specifically to buy Luo Tingzhu another pair of shoes, and then bought himself another pair.

There was no other way; the teacher had arrived, and his slippers were retired. For a while, he would just wear whatever he could find when entering the lab. Now that the student was in the lab, he could wear Fang Ziye's slippers.

Fang Ziye accidentally slipped Luo Tingzhu's shoes on his feet again. Yuan Weihong returned the shoes, and Fang Ziye didn't mind. But if he returned the slippers, the girl might not think the same way.

So Fang Ziye simply threw away Luo Tingzhu's slippers and bought two new pairs.

They lost Luo Tingzhu's shoes.

"Do you really know it, or are you just pretending? These preliminary experiments are for the research grant application, so there can't be any mistakes. If you don't know it, what will you do?" Yuan Weihong reminded Fang Ziye just before he was about to begin.

"Master, why don't you go back first and come back to check when it's time?" Fang Ziye wanted to make up for his previous mistake and wanted his teacher, Yuan Weihong, to rest.

In typical Transwell experiments, the inner chamber is usually referred to as the upper chamber, and the inner chamber of the cell culture plate is referred to as the lower chamber. The bottom of the chamber is a polycarbonate microporous membrane, on which a layer of Matrigel is laid.

Matric gum is a matrix component extracted from mouse sarcomas rich in extracellular matrix proteins. Its main components include laminin, type IV collagen, nestin, and many growth factors. Matric gum coagulates at room temperature into a biologically active three-dimensional matrix that can mimic the extracellular matrix. Similar to the in vivo metastasis of tumor cells, invasive cells secrete matrix metalloproteinases to break down the matric gum and further metastasize to the outside.

Fang Ziye has now completed the dilution of serum-free culture medium and matrix gel, and is currently performing an ice bath procedure.

This procedure involves diluting the matrix gel at a ratio of approximately 1:8 using serum-free medium on ice, adding 100 μL/well to the upper chamber, covering the bottom, and incubating in a cell culture incubator for 1-4 hours.

There's no need for Yuan Weihong to wait here during this waiting period.

But for the operation four hours later, Fang Ziye and Luo Tingzhu would need to wait here. It was still relatively early.

The layman sees the details, the expert sees the spectacle. Seeing that Fang Ziye's operation was quite skillful, Yuan Weihong looked around and nodded.

He said, "You need to be a little more careful when you do things."

"If you don't understand something, learn from your junior sister. Ask more questions; asking more questions won't cost you anything," Yuan Weihong advised before walking away with his hands behind his back.

As soon as Yuan Weihong left, Luo Tingzhu turned around nervously and asked, "Senior Brother Fang, have you been in the lab before? How come you're so skilled at these operations?"

As Yuan Weihong said, these experiments were preliminary experiments for her research grant application. The funding was limited, and she couldn't afford too many failures; otherwise, she wouldn't have asked Professor Yuan Weihong for help.

However, Luo Tingzhu never expected that Fang Ziye, who was even more of a novice than herself, would be so skilled.

But he's probably just a newbie on the surface; it's possible Fang Ziye secretly went into the lab before. After all, he's a third-year master's student and has been in orthopedics for so long.

Every operation involves a process from knowing how to do it to becoming unfamiliar with it, and then to becoming proficient.

Fang Ziye looked at the 4*6 24-well culture plate and saw no major problems. He replied, "I've looked at it a few times, but it should be fine. Junior Sister Luo, let's go to the training room while we have this time."

“Waiting here isn’t going to solve anything. Your appointment for today’s operation is going to take a while, right?” Fang Ziye asked.

Both the operating table and the cell operation room require reservations.

Luo Tingzhu nodded; she was definitely taking her own affairs to heart.

“Then there’s no problem. Let’s go to the practice room together. Four hours later, come back and add serum-free culture medium, then add complete culture medium, and then continue culturing for another 24-48 hours.” Fang Ziye knew this by heart.

It was the success of the previous scratch test that gave Fang Ziye a certain degree of confidence.

"Okay!" Seeing that Fang Ziye wasn't worried, Luo Tingzhu felt a little relieved.

Fang Ziye wasn't a reckless person; his meticulousness was no less than hers. When Luo Tingzhu secretly took photos for Fang Ziye before, it was just something she did on a whim, and she didn't dare remind him.

After all, Fang Ziye's own timeline was based on articles for reference. Standardized recording and photography were merely for obtaining more experimental material and had no particular significance, so they wouldn't appear in the articles.

If there were supporting literature, the editorial department probably wouldn't have asked for images from time points that weren't relevant. It's just that Yuan Weihong had high expectations for Fang Ziye, which is why the requirements were a bit higher.

As they walked towards the practice room, Luo Tingzhu asked with some concern, "Senior Brother Fang, will you apply for school-level and city-level research projects?"

Fang Ziye is also a master's student and a student at Han City University.

Luo Tingzhu is eligible to apply, and Fang Ziye's qualifications are in no way inferior. Moreover, Fang Ziye has a foundation in basic-type articles, making him more capable. In addition, his operational proficiency is higher, allowing him to conduct better preliminary experiments.

If it were a direct confrontation, Luo Tingzhu estimated that she might not be able to defeat Fang Ziye, and only hoped that Fang Ziye could choose one of them.

"I'll probably give it a try and do some research during my doctoral studies," Fang Ziye replied sincerely. But seeing Luo Tingzhu's expression change slightly, Fang Ziye added, "Junior Sister Luo, as long as the quality of the research is good enough, there won't be any problem."

"There is no rule that the foundation can only fund one orthopedic-related project."

Fang Ziye knew that Luo Tingzhu desperately needed funding to conduct her experiments; only by successfully completing the experiments and publishing papers could she possibly remain in the hospital.

Luo Tingzhu smiled and shook her head at Fang Ziye. She seemed very composed, but her body shrank slightly, whether out of fear or a subconscious defensiveness, it was hard to tell.

Even after seeing Luo Tingzhu's reaction, Fang Ziye's heart remained largely unmoved.

Luo Tingzhu needs research funding to conduct experiments, and Fang Ziye also needs research experience to build his resume. They are all ordinary people struggling to make ends meet. Who would dare to show pity and say, "What opportunity will I give you?"
Luo Tingzhu remained silent the entire way. After entering the training room, she told Fang Ziye that she would go practice first, and then the two took their places.
